HIGHLIGHTS
Kisspeptin-10 (also known as Metastin 45-54) is a potent decapeptide that serves as the "master switch" for the mammalian reproductive system. It is the minimal active fragment of the larger Kisspeptin-54 protein and acts as a high-affinity endogenous ligand for the GPR54 (KISS1R) receptor. In research models, Kisspeptin-10 is studied for its unique ability to directly stimulate GnRH (Gonadotropin-Releasing Hormone) neurons in the hypothalamus. This trigger initiates a natural, pulsatile cascade that increases the secretion of Luteinizing Hormone (LH) and Follicle-Stimulating Hormone (FSH) from the pituitary gland, subsequently driving the production of endogenous sex steroids.
Observed Benefits
-
HPG Axis Activation: Restores or enhances the Hypothalamic-Pituitary-Gonadal axis by mimicking natural GnRH triggers.
-
LH/FSH Stimulation: Highly effective at inducing rapid, dose-dependent rises in Luteinizing Hormone, essential for studying testosterone production and ovulation.
-
Metastasis Suppression: Originally discovered as "Metastin," it is heavily researched for its ability to inhibit the migration and spread of certain cancer cell lines.
-
High Specificity: Unlike direct GnRH agonists, Kisspeptin-10 works "upstream," potentially allowing for a more physiological stimulation of the endocrine system.
-
Rapid Action: Characterized by a fast onset of action, making it ideal for acute research observations of hormonal pulses.
Protocol Overview
Kisspeptin-10 research typically involves subcutaneous administration. To maintain a standardized laboratory environment at Biolabs Peptides South Africa, we utilize 3.0 mL of Bacteriostatic Water per 10mg vial. This creates a concentration of approximately 3.33 mg (3333 mcg) per mL. Because Kisspeptin-10 has a relatively short half-life (~4 minutes in blood), research protocols often focus on acute pulse stimulation or twice-daily administration to maintain elevated receptor activity.
Research Protocol Dosing
The following measurements provide the required units on a U-100 insulin syringe based on the 3.0 mL reconstitution.
Phase 1: Initial Pulse Assessment (100 mcg per dose)
-
Purpose: To observe initial pituitary sensitivity and baseline LH response.
-
Measurement: 3 units (0.03 mL) on a U-100 syringe.
Phase 2: Standard Research Dose (200 mcg to 300 mcg per dose)
-
Purpose: Studied for significant stimulation of the HPG axis and testosterone/estrogen modulation.
-
200 mcg Measurement: 6 units (0.06 mL) on a U-100 syringe.
-
300 mcg Measurement: 9 units (0.09 mL) on a U-100 syringe.
Phase 3: High-Intensity Stimulation (500 mcg per dose)
-
Purpose: Investigating maximal receptor occupancy and acute gonadotropin spikes.
-
Measurement: 15 units (0.15 mL) on a U-100 syringe.
Frequency: Research typically involves 1 to 2 administrations daily. Prolonged continuous use may lead to receptor desensitization (tachyphylaxis), so cycles are generally limited to 2–4 weeks followed by an equal rest period.
Supplies Needed
-
Biolabs Peptides Kisspeptin-10: 10mg Lyophilized Vial.
-
Reconstitution Liquid: 3.0 mL of Bacteriostatic Water.
-
Syringes: U-100 Precision Insulin Syringes (0.3 mL or 0.5 mL capacity for high accuracy).
-
Prep: Medical-grade alcohol swabs for vial and site prep.
Storage
-
Lyophilized State: Store at -20°C (-4°F) for long-term stability (up to 24 months). Protect from UV light.
-
Reconstituted State: Must be refrigerated at 2–8°C (36–46°F). Kisspeptin-10 is a delicate decapeptide; once in solution, it is best used within 14–21 days to ensure peak bioactivity.
-
Handling: Do not shake the vial. Gently roll or swirl until the solution is clear.
