Tesamorelin is a synthetic analogue of the Growth Hormone-Releasing Hormone (GHRH). It is a stabilized 44-amino acid peptide that acts specifically on the pituitary gland to stimulate the natural, pulsatile secretion of endogenous Growth Hormone (GH). In research models, Tesamorelin is highly regarded for its unique ability to target and reduce visceral adipose tissue (VAT) without significantly impacting subcutaneous fat or glucose metabolism. Unlike direct GH administration, Tesamorelin preserves the natural feedback loop of the HPA axis, making it a primary focus for studies involving metabolic syndrome, lipodystrophy, and age-related GH decline.

Protocol Overview

Research involving Tesamorelin utilizes daily subcutaneous administration, typically performed in a fasted state to maximize pituitary response. To ensure standardized and accurate results in South African laboratory environments, each 10mg vial is reconstituted with 3.0 mL of Bacteriostatic Water. This creates a concentration of approximately 3.33 mg (3333 mcg) per mL.

Research Protocol Dosing

Concentration Reference:

  • 10mg Vial + 3.0 mL: ~3.33 mg per mL (1 unit = ~33.3 mcg).

The following measurements provide the required units on a U-100 insulin syringe for a daily research schedule. Tesamorelin is typically administered 5 to 7 days per week.

Phase 1: Initial Assessment (500 mcg Daily)

  • Measurement: 15 units on a U-100 syringe.

  • Purpose: To observe initial pituitary sensitivity and baseline IGF-1 response.

Phase 2: Standard Research Dose (1,000 mcg / 1 mg Daily)

  • Measurement: 30 units on a U-100 syringe.

  • Purpose: The most common dose studied for metabolic regulation and visceral fat reduction

Phase 3: High-Intensity Research (2,000 mcg / 2 mg Daily)

  • Measurement: 60 units on a U-100 syringe.

  • Purpose: Studied in clinical trials for maximum efficacy in treating severe lipodystrophy.

Frequency: Administration is best conducted once daily, ideally in the evening or immediately upon waking, at least 90 minutes away from food.

Supplies Needed

  • Biolabs Peptides Tesamorelin: 10mg Lyophilized Vial.

  • Reconstitution Liquid: 3.0 mL of Bacteriostatic Water.

  • Syringes: U-100 Precision Insulin Syringes (0.5 mL or 1.0 mL recommended).

  • Prep: Sterile alcohol swabs for vial and site sterilization.

Storage

  • Pre-reconstitution (Lyophilized Powder): Store frozen at -20°C (-4°F) for long-term stability (up to 24 months). Protect from UV light.

  • Post-reconstitution (Liquid Solution): Must be kept refrigerated at 2–8°C (36–46°F). Once mixed, the solution should be used within 14–21 days for peak bioactivity.

  • Handling: Tesamorelin is a large, fragile peptide. Do not shake the vial. Gently roll or swirl until the powder is fully dissolved and the solution is clear.
