How to Reconstitute Peptides: A Simple Step-by-Step Guide

Correct reconstitution is one of the most important steps in working with peptides. Even the highest-quality, lab-grade peptide can lose effectiveness if it’s mixed or handled incorrectly.

This guide explains how to reconstitute peptides safely and correctly, using best-practice handling methods recommended by laboratories and peptide suppliers worldwide.

What Does “Reconstitution” Mean?

Most peptides are supplied as a lyophilised (freeze-dried) powder. Reconstitution simply means adding a sterile liquid — typically bacteriostatic water — to turn the powder into a usable solution.

This process must be done carefully to:

  • Preserve peptide integrity

  • Avoid contamination

  • Ensure accurate dosing

  • Extend shelf life after mixing

What You’ll Need

Before starting, make sure you have the following supplies ready:

• Peptide vial• Bacteriostatic water• Syringe• Alcohol wipes

Work on a clean, sanitised surface and wash your hands thoroughly.

Step-by-Step: How to Reconstitute Peptides

Step 1: Open the Vial Tops

Carefully remove the protective plastic caps from both the peptide vial and the bacteriostatic water vial.Do not remove the rubber stoppers.

Step 2: Sanitise

Use alcohol wipes to clean the rubber stoppers on both vials.Allow them to air dry for a few seconds before proceeding.

Step 3: Draw the Water

Insert the syringe into the bacteriostatic water vial, hold it upside down, and draw the required amount of water (in millilitres) into the syringe.

The amount of water added depends on how concentrated you want the final solution to be.

Step 4: Inject the Water

Hold the peptide vial slightly at an angle and slowly inject the water down the side of the vial, not directly onto the powder.

This helps prevent foaming and allows the peptide to dissolve more evenly.

Step 5: Dissolve the Peptide

Gently roll the vial between your fingers for 5–10 seconds.Do not shake aggressively.

Place the vial in the fridge for approximately 15 minutes, allowing the powder to fully dissolve.

Step 6: Check the Solution

Once fully reconstituted, the solution should appear:

  • Clear

  • Colourless

  • Free of particles or cloudiness

If the solution looks cloudy or gelled, do not use it.

Does Shaking Damage Peptides?

No.The idea that shaking peptides destroys them is an old myth. Independent testing laboratories — including Janoshik Analytical — have shown that shaking does not cause degradation or loss of potency.

That said, gentle handling is still recommended for consistency and ease of dissolution.

How Should Reconstituted Peptides Be Stored?

• Store reconstituted peptides in the fridge (2–8 °C)• Ideally use within 4 weeks• Protect from light (store in a pen or light-blocking container)• Avoid unnecessary temperature changes

Lyophilised (unmixed) peptides can be stored longer in the fridge or freezer, depending on intended use timeframe.

Why Use Bacteriostatic Water?

Bacteriostatic water contains 0.9% benzyl alcohol, which prevents bacterial growth. This allows reconstituted peptides to remain sterile for up to 28 days, making it safer and more practical than sterile water for multi-dose use.

Sterile water contains no preservative and is generally intended for single-use applications.

What If Something Doesn’t Look Right?

If your peptide solution appears:

  • Cloudy

  • Gelled

  • Particulate
