Retatrutide 5 mg Vial: Dosage & Titration Protocol

Retatrutide is a potent "triple-agonist" peptide. To minimize side effects like nausea or heart rate fluctuations, a slow titration (dose escalation) schedule is necessary.

1. Reconstitution (Mixing)

  • Diluent: Add 1.0 mL of Bacteriostatic Water to your 5 mg vial.

  • Concentration: This creates a 5 mg/mL solution.

  • Measurement: On a standard U-100 (1 mL) insulin syringe:

    • 10 Units = 0.5 mg

    • 20 Units = 1.0 mg

    • 40 Units = 2.0 mg

2. The Titration Schedule

(Doses are injected once per week)

Phase 1: Initiation (Weeks 1 through 4)

  • Weekly Dose: 2 mg

  • Syringe Volume: 40 Units

Phase 2: Escalation (Weeks 5 through 8)

  • Weekly Dose: 4 mg

  • Syringe Volume: 80 Units

Phase 3: Maintenance (Weeks 9 through 12)

  • Weekly Dose: 6 mg

  • Syringe Volume: 120 Units (Note: Since this exceeds a 100-unit syringe, you will need to draw two separate injections, e.g., 60 units twice, or use a second vial).

Phase 4: Maximum Dose (Weeks 13 and beyond)

  • Weekly Dose: 8 mg

  • Syringe Volume: 160 Units (Requires multiple injections).

3. Administration & Storage

  • Method: Subcutaneous injection into the abdomen, thigh, or upper arm. Rotate sites weekly.

  • Frequency: Once every 7 days.

  • Storage: Keep unopened vials in the freezer. Once mixed (reconstituted), store in the refrigerator and use within 28 days.

4. Side Effects & Support

  • Common Effects: Nausea, mild diarrhea, or appetite suppression.

  • Adjustment: If side effects are difficult to manage, do not increase the dose. Stay at the current level for another 4 weeks until your body adjusts.

  • Hydration: Drink plenty of water and prioritize protein intake.
